چکیده

MOTIVATION Microarrays are being increasingly used in cancer research to better characterize and classify tumors by selecting marker genes. However, as very few of these genes have been validated as predictive biomarkers so far, it is mostly conventional clinical and pathological factors that are being used as prognostic indicators of clinical course. Combining clinical data with gene expression data may add valuable information, but it is a challenging task due to their categorical versus continuous characteristics. We have further developed the mixture of experts (ME) methodology, a promising approach to tackle complex non-linear problems. Several variants are proposed in integrative ME as well as the inclusion of various gene selection methods to select a hybrid signature. RESULTS We show on three cancer studies that prediction accuracy can be improved when combining both types of variables. Furthermore, the selected genes were found to be of high relevance and can be considered as potential biomarkers for the prognostic selection of cancer therapy. AVAILABILITY Integrative ME is implemented in the R package integrativeME (http://cran.r-project.org/).

Metformin and Pioglitazone Reduce Gene Expression of Inflammatory Factors in Insulin Resistant and Hypertrophied Adipocytes

Objective: In obesity, chronic low grade inflammation, created by induction of pro-inflammatory markers, causes adipocyte dysfunction in adipose tissue. Adipocytes dysfunction is associated with various diseases including insulin resistance and obesity. In obesity, inflammatory factors such as osteopontin (OPN), angiopoietin-like protein 2 (Angptl2) and transforming growth factor-β (TGF-β) are...

Simple procedure for production of short DNA size markers of 100 to 2000 bp

DNA size markers (ladder) are essential tools in molecular biology, genetics and biotechnology. In this study, a simple and cost-effective method for laboratory production of DNA ladders is introduced. For this purpose, different sizes of 100 to 2000 bp DNA segments were designed using PCR technique. For producing 14 different gene fragments as DNA molecular weight markers, recombinant plasmid ...

افزایش نرخ کارایی طبقه بندی با استفاده از تجمیع ویژگی های موثر روش های مختلف ترکیب شبکه های عصبی

Both theoretical and experimental studies have shown that combining accurate Neural Networks (NN) in the ensemble with negative error correlation greatly improves their generalization abilities. Negative Correlation Learning (NCL) and Mixture of Experts (ME), two popular combining methods, each employ different special error functions for the simultaneous training of NN experts to produce negat...

Integrative vectors for gene deletion and replacement.

An improved method for gene deletion or replacement in Escherichia coli was developed. It employs a set of integrative vectors and two helper plasmids, as a temporary source of RecA and Flp activities. The integrative vectors combine several useful features including three different selection markers placed between two parallel oriented Flp recombinase target (FRT) sites. Each marker is flanked...

Construction of Recombinant Vectors Containing Clavulanic Acid Antibiotic Regulatory Gene, claR, Isolated of Streptomyces clavuligerus Strains

The claR of Streptomyces clavuligerus in the clavulanic acid gene cluster encodes a transcriptional regulator that controls clavulanic acid biosynthesis. The main goal of this study was isolation and molecular detection of the claR gene and its cloning in the Streptomyces specific vector (pMA:: hyg). By cinsideration of the claR gene’s start codon, the specific primers were designed. After geno...
